Blood testing is one of the most common methods for measuring physiological indicators and assessing human healthFor a clearer grasp of this concept, let’s review three important blood-related definitions and know more about its key components:

  1. od: A mixture of blood cells (including red blood cells, white blood cells, and platelets), proteins (such as albumin, globulin, fibrinogen, etc.), glucose, mineral ions, hormones, and waste products.
  2. Plasma: The liquid portion of blood containing all components except blood cells.
  3. Serum: Obtained by removing blood cells and certain proteins (like fibrinogen) .the fibrinogen proteins of blood  form clots by binding with blood cells, and serum is the result of separation of these clots from blood.

 Serum, the optimal medium for Diagnostic test:

 Most diagnosis tests are performed on blood serum due to its:

  1. limited interfere with test results
  2. stable composition which remains consistent over time compared to other blood components

Thus, developing an effective method to isolate serum—without altering its integrity and ensuring complete separation until testing—is critical.

 The Separation Process

After blood collected in test tubes, samples are left to clot . The samples are then centrifuged to separate the clot from the serum.

 How SSG works

The SSG is pre -injected into blood collection tubes during assembly, where it settles at the bottom of the tube. During centrifuge process, the SSG moves from the bottom and positions itself at the interface between the serum and clotted blood. the result impervious layer, with serum collected above it , enables  clean serum extraction for diagnostic tests.

Key Requirements for an Effective SSG

1- Density: intermediate between serum and blood cells

2- Inertness: must neither absorb nor release substances into the serum, and must not chemically interact with it

3- Sterilization resistance: Must withstand gamma radiation (used for tube sterilization)

4- Efficiency : Should achieve separation at low centrifuge speed and short duration
